Error loading OS...

First of all, thanks in advance to all that can help me.
Here's my problem with my desktop. I usually don't turn the computer off and just
put it in sleep mode until I have to use it again. So far, I've never had a problem
when doing this. Recently I did turn the entire computer off because we took a trip
out of state. Now I go to turn the computer back on, and all I get is a black screen
with a message "error loading OS." I had all the updated antivirus definitions, and
two good firewalls on this computer so I'm kinda lost as to what the problem could
be or how to fix it. Any and all help is greatly appreciated.
