Rather jumping around from server to the router, few troubleshooting steps i would
take would be to disable services and task which you don't need immediately e.g antivirus,
schedule tasks and anything else you might be running updates and stuff. basically
strip the server to bare minimum. I would start by checking the events on the server
during the time period i would also turn on all the audit policies within the GPedit
so you can see what service is starting or stopping during the time period above.
have you rebooted the server rather then clients? and what happens does it resolve
the problem. To me it seems like something is hogging bandwidth i would run bandwidth
check. Also i would look at server performance counters. check the logs on the rounters
see what you can come up with .good luck hope my 2 cents help
