The USB port is connected to the computer, not to the hard drive inside the computer. The answer is no, and wasn't really simple. Adding internal drives is pretty easy, if you wanted to pull the old PC's drive out of it.

Each X86 computer starts out naming its primary drive's "DOS Partition" its C: drive, but there is nothing really permanent about that, and you can mix and match drives quite readily, without running into conflicts that you can't easily overcome.
