Start -> Run -> regedit, delete the key:
HKEY_CURRENT_USER\Software\Microsoft\Windows\CurrentVersion\Policies\System
Press F5 and see if it comes back. If yes or if you can't run regedit, then you have some malware running. Since disabling tools like TaskManager is a common way for malwares to protect themselves, run some tools to make sure you don't have one. A replacement for TaskManager that cannot be disabled this way can be downloaded here.
