Bounced Mails: Error 550 5.7.1

For past few days, a number of e-mails from outside addresses (like Yahoo! and others) 
to our organisation mail addresses are being bounced. The bounced message comes from 
 Postmaster@... (our ISP) and the error is: 

Mailbox unknown or not accepting mail. 
550 5.7.1 Unable to relay for xxx@... (our organisation mail address) 

Around one month back, we had installed a LAN Proxy Server (Squid). After installing 
that, our internet connectivity has improved but the mail problems have started. 
I wonder if Squid Proxy is anyway connected with relaying of mails or there is some 
problem at our ISP end.

Can you please suggest suitable diagniostics and remedies?
